The idea that the OP had would show good humor, I agree, but in all
likelihood few people on the receiving end would 'get' it, in the least. I'd
say that you would be best served by presenting only direct communication
products (i.e., the cover letter and resume are communication products) to
them and save the valuable insight, humor, and good attitude for in-person
interviews.

The 'teaser hook' idea, as mentioned below, can be a good one but I believe
that should you decide to use such, in your cover letter, then you should
write it in such a clear way that anyone reading it there would immediately
recognize it.

an ISO process overview with work instructions.

If I was a manager looking at cover letters and resumes, this would get my
attention and would make me want to see that portfolio.

Unless they're strictly "corporate" who doesn't want any employees with
traits like humor and passion -- in which case we would not be happy anyway
so it's a good thing our time does not get wasted in a useless interview.
