So the question is, then, how to readers act? I don't know the answer to
that one.
In that case,m wouldn't it be best to put every glossary term in the index - suitably identified, e.g.
term 45, 128-134
explained 322
That way, you'd accommodate both readers who are aware of a glossary and use it and those who aren't.
